TL;DR
The fastest public-transport option from IST is usually the M11 metro to Gayrettepe, followed by M2 or tram/funicular connections. With luggage, Havaist airport buses are often easier: they serve Taksim and the Aksaray/Beyazıt area for onward travel to Sultanahmet.
For Taksim, take a direct Havaist airport bus to the Taksim stop; it is convenient with luggage, although travel time depends on traffic. Alternatively, take M11 to Gayrettepe and transfer to the M2 metro towards Taksim.
For Karaköy, take M11 from the airport to Gayrettepe, then M2 to Şişhane. From Şişhane, walk downhill or use the F2 funicular via Tünel; with heavy luggage, a Havaist bus to Taksim plus an onward connection may be easier.
For Sultanahmet, a practical public-transport route is a Havaist bus towards Aksaray/Beyazıt, then the T1 tram towards Kabataş, getting off at Sultanahmet. The M11 metro does not reach the Old City directly, so it requires multiple transfers.
From Fatih and the Marmaray line, first connect to the M11 interchange, typically by using central metro lines to Gayrettepe. An Istanbulkart balance of around 300 TL should normally cover several metro trips with transfers, but check current Havaist and transit fares before travelling.
