Best to visit Nusa Penida with an overnight stay: day one for island beaches, day two for manta snorkeling at Manta Point (about 3 hours by boat). For diving with mantas it's easier to go from Sanur, for snorkeling — from Nusa Penida itself; chances of seeing mantas are higher at Manta Point than Manta Bay.
There are two spots for manta encounters: Manta Bay and Manta Point. Manta Bay is closer and usually included in one-day tours, but chances of seeing mantas there are lower. Manta Point is better — for snorkeling it's easier to reach from Nusa Penida itself, while for diving it's easier from Sanur. A one-day island tour plus a separate diving day at Manta Point is usually enough to minimize transfers. Snorkeling around Nusa Penida itself is also beautiful, especially at Crystal Bay — worth staying longer on the island just for that.
Manta Point is located on Nusa Penida island itself. There are usually no direct boats from Sanur port straight to snorkeling — snorkeling trips use small open boats for 15-20 people, while boats from Sanur to the island are large closed ferries carrying about 100 people. If you want to combine sightseeing and manta snorkeling, it's best to stay overnight: day one for the island's beaches, next morning for Manta Point snorkeling (about 3 hours). You can arrange a private driver in advance who will plan the route, organize the snorkeling trip, and even help book cheaper boat tickets from Sanur than through travel agencies.
